LA - English UL - http://discoverylib.upm.edu.my/discovery/Record/422006 AB - A cytogenetic study was conducted on the freshwater prawns, Macrobrachium rosenbergii, locally known as udang galah. They were obtained from Linggi river, Negeri Sembilan. Metaphase chromosome spreads were prepared from gill tissues and tissues of testes from adult and juvenile prawns and also from tissues of whole body parts of nauplis larvae. Ten adult prawns with mean body weight of 65.0±25.0 and mean total length of 25.0±15.0cm, 30 juveniles prawn with mean body weight of 3.0±0.1g and mean total length of 5.0±1.0cm, and 300 larvae with mean total length of 1.0mm were used based on four techniques. In the first technique, tissue of gills from 5 adults and 10 juvenile prawns were used after intramuscular injection of Lee's solution with different duration of treatments. In the second technique, tissues of gill from 10 juvenile prawns were used after treatment in 8 hours of Lee's solution and later dipped in hypotonic solution (ddH2O) for 45 minutes Tissue of testes from 5 adult prawns and 10 juvenile prawns were used after treatment in 0.005% colchicine solution. For the nauplius larvae, the whole body tissues were used after treatment in 0.005% colchicine solution. Over 500 slides were observed for chromosome spread using light compound microscope. Only the second technique using gill tissues of the juvenile prawns treated in Lee's solution with modification showed chromosome spreads. The good spread of metaphase chromosomes could not be obtained for chromosomes counting of the Macrobrachium rosenbergii due to overlapping of chromosomes.
