Male Sprague-Dawley rats (n =40) weight 200g to 300g were divided into six groups (n =8): which consisted of normal control group (NC), diabetic control group (DC), diabetic treated with 100mg/kg body weight of pomegranate seed (DPS), diabetic treated with 100 mg/kg body weight of aril (DPA), diabetic treated with 100 mg/kg body weight of pomegranate juice (DPJ) and diabetic treated with 10mg/kg body weight of glibenclamide (DG), an anti-diabetic drug. Pomegranate seed and aril was prepared by freeze dried the seed and aril, grinded into powder from, and mixed with distilled water. Pomegranate juices was prepared by squeezed pomegranate aril, filtered juice and diluted with distilled water. The pomegranate seed, aril and juice was administered into rats through force feed method for 28 days. The body weight measurement and blood glucose level screening via glucometer were carried out on day 0, 14 and 28. The blood samples for insulin level analysis using enzyme linked immunosorbent assays (ELISA) method were collected via cardiac puncture on day 28. On day 28, the study was terminated and liver was isolated for histopathology study. The results showed significant increment (p <0.05) of mean body weight for DP) group after treatment. DPA group showed reduction on mean body weight, but result was not significant. For blood glucose level, all groups showed elevation of blood glucose level after treatment. Least elevation was showed in DPA group, but the result was not significant. DPJ group showed significantly increment p <0.05) of mean blood glucose level after treatment. For insulin level, no significant effect was shown in regulating insulin level for all groups. DPJ groups showed highest insulin level followed by DPA group, DG group and DPS group, yet all result was not significant. Besides, there was no observable difference of liver histology between groups. In conclusion, 100 mg/kg body weight of pomegranate aril was able to reduce body weight, but did not change blood glucose level significantly. Furthermore, pomegranate did not pose toxicity effect on liver. Longer study period may be needed to investigate the effects of pomegranate thoroughly in future studies. CN - FPSK3 2013 48 ER -
