| Riassunto: | Mammalian cells are constantly exposed to a wide variety of genotoxic agents. In several DNA repair genes, polymorphism may result in reduced repair capacity, which has been implicated as a risk factor for various types of diseases. The frequency of the polymorphic alleles varies among populations. The X-ray repair cross-complementing group 3 (XRCC3) belongs to a family of genes responsible for repairing DNA double strand breaks caused by normal metabolic processes and/or exposure to ionizing radiation. Polymorphism of XRCC3 gene has a sequence variation in exon 7, which cause C to T variant. This will result in an amino acid change at codon 241 (Thr241Met) of DNA repair gene XRCC3. The objective of this study was to determine the genotype and allele frequencies of XRCC3 T241M in a student population. A total number of 124 samples were collected from buccal epithelial cells using Cytobrush. Genomic DNA was extracted from buccal epithelial cells using the DNA extraction kit. Genotyping of codon 241 of XRCC3 gene was done by PCR restriction fragment length polymorphism (RFLP), using NlaIII restriction enzyme and was resolved on agarose gel. The genotype frequencies of XRCC3 gene were as follows; 81.45% TT, 17.74% TM and 0.81% MM. The allele frequencies were 0.9 and 0.1 for Thr and Met alleles of XRCC3 T241M. This finding suggests that T241M genetic polymorphism of XRCC3 gene is normally distributed in student population. |
