There are many research on P. hypophthalmus for the growth, habitat and stocking density. However, the study of the reproductive system is less compared to the other study on P. hypophthalmus. This study involved detection and characterization of the gonadotropin (GTH) subunits which are α, Follicle Stimulating Hormone (FSH) and Luteinizing Hormone (LH) in different organs such as pituitary, brain, kidney, gonad and spleen that have involved in reproduction system of fish. The first method that used in this study was collection of organs sample from the UPM hacthery located in Puchong, Selangor. Then, the organs sample was extracted for the RNA. Next, RNA was transcribed into cDNA which used for amplification by Polymerase Chain Reaction (PCR). The PCR amplification was used to detect the present of GTH subunit (α, FSHβ and LHβ) in organ samples. Later, PCR was used to characterize the GTH subunit. The result for molecular detection in different organs showed that α subunit located in pituitary with 180 bp, FSHβ also located in pituitary with 120 bp and LHβ located in gonad with 151 bp. For the molecular characterization result, PCR successfully amplified α with 443 bp, FSHβ with 120 bp and 513 bp, respectively. The similarity between other fish species showed that α subunit have the similarity with Channel catfish (Ictalurus punctatus) with percentage value 95% similarity, FSHβ have similarity with North African catfish (Clarias garienpinus) with percentage value 94% and LHβ have the similarity with Malaysia river catfish (Hemibagrus nemurus) with the percentage value 98%. Phylogenetic tree showed that P. hypophthalmus have highest similairy with its own order which is Siluriformes, followed by Cypriniformes. This showed that P. hypophthalmus is depending on their order and species. Result of this study may help in the contribution of gene expression and production of recombinant hormone to improve the production of P. hypophthalmus and related species for aquaculture industry in Malaysia. | ||
