520 0 |a Breast cancer is a major malignancy among women worldwide. This malignancy is the main cause of cancer-deaths in women.In Malaysia, 1 in 16 women have a lifetime risk of breast cancer. Cell lines are widely used as in vitro models in cancer research to identity key processes involved in the initiation and progression of cancer, as well as the efficacy prediction of novel theraupeutic compounds. However, breast cancer cell lines currently used are mostly derived from the Caucasian population. The major objective of this project was to establish immortalized Malaysian primary breast cancer cell lines to facilitate the creation of a Malaysian breast cancer model. This project comprised of 4 major methodological parts. Immortalization of breast cancer cells from the malignant breast cancer specimens was the first part of the project. After immortalization, the cells were genotypical characterization by PCR-based mRNA expression profiling. Total RNA was extracted from the cells and reversed-transcribed into cDNA before PCR was conducted to evaluate the expression of estrogen receptor (ERα) and progesterone receptor (PR). Protein quantification of the relevant proteins was executed using western blot. Finally, the cell lines were characterized phenotypically. MCF7 cells were used as a control. After subsequent passages, two cell lines, C1 and C2 were created, whereby the cells proliferated consistently and apparently infinitely. This observation suggests that immortalization was achieved. C1 was ERα and PR positive ehile C2 was ERα positive but PR negative. Wound healing and proliferation assays were demonstrated in each of the cell lines for 24 hours and 48 hours respectively. In conclusion, this is to date the first isolation and establishment of immortalized Malaysian primary breast cancer cell lines.
