Haruan (Channa striatus) traditional extract (HTE) traces by gamma camera using technetium (99mTc). /

Haruan (Channa striatus) has been shown containing high concentration of glycine, the most important component of human skin collagen and high level of arachidonic acid. These compound has potent anti-inflammatory and analgesic activities since few studies had demonstrated that a lipoamino acid, ara...

These compound has potent anti-inflammatory and analgesic activities since few studies had demonstrated that a lipoamino acid, arachidonoylycine suppressed oedema and pain. Haruan extract contains high arachidonic acid, precursor of prostaglandin thus initiate blood clotting and play potential role in wound healing. This study is done to trace haruan extract (HTE) in the circulation system of rats by viewing it under gamma camera. By understanding it, the delivery and distribution of extract can be viewed in the circulation system. Rats underwent metabolic turnover rate study, where they were given diets of 0%, 10%, 20% and 25% extract in the pellet ad libitum for seven days. The rats were weighed daily. The metabolic rate of rats increases significantly (P<0.05) with the increase of body mass. At the end of seven days, the metabolic rate for rat taking regular pellet, 10% extract, 20% extract and 25% extract were 15.791±0.691, 18.609±0.971, 0.192±0.006 and 20.569±0.685 kCal respectively. Haruan extract was radiolabeled with technetium (99mTc) and underwent quality control before used. The labeling efficiency of haruan extract labeled with 99mTc is 27.5%. After purification using PD-10 size exclusion, the radiolabelling efficiency was 94.67%. The fraction was collected after 300 µl of 0.9% NaCl, at the 7th and 8th fraction. This purified fraction was 130.1 µCi respectively. Rats were anesthetized with 5mg/kg Ketamine/Xylazine intraperitoneally before injected with radiolabelled haruan extract intravenously. Gamma images were taken every 30 seconds for 40 minutes for the dynamic study. For static study, gamma images were taken three hours after injection. The radiolabeled extract was traced in the liver, lungs, heart, kidney and urinary bladder after 40 minutes. After three hours, almost all of the extract was excreted into urinary bladder.
